Inhibition of Nitric Oxide Synthase by Alpinia officinarum

Abstract
Nitric Oxide (NO), derived from L-arginine, is produced by two types of nitric oxide synthase (constitutive NOS: cNOS, inducible NOS: iNOS). The N0 produced in large amounts by the iNOS is blown to be responsible for the vasodilation and hypotension observed in septic shock and inflammation. The inhibitors of iNOS, thus, may be useful candidates for the treatment of inflammatory diseases accompanied by the overproduction of NO. The activity guided column chromatography of the chloroform soluble fraction obtained from the methanol extract of Alpinia officinarum gave an inhibitory active compound of NO synthesis in LPS-activated macrophages (IC50 value; 0.6¥ìM). The structure of active compound was elucidated as 7-(4-hydoxy-3-methoxyphenyl)-1-phenylhept-4-en-3-one by spectral analysis. This compound can be an useful candidate leer the development of new drug to treat endotoxemia and inflammation accompanied by the overproduction of NO.
